A refractive index mismatch between the oil immersion and the microscopic object can lead to a severe over-estimation of the object's size. The cause of this effect is explained and a simple calibration method to compensate for its occurrence is presented. A practical example is discussed. The analysis applies to both conventional three-dimensional, and confocal microscopy.
