METHODSDogs weighing between 12 and 31 kg and not selected for sex or breed were anesthetized with a combination of morphine sulfate, 10 rng/kg, SC, followed in 30 min by pentobarbital sodium, 15 mg/kg, iv. A tracheal cannula was inserted and a piston-type air pump was used to control respiration. Both vagus nerves were isolated in the neck and cut to reduce reflex changes of intrinsic vasomotor activity when arterial pressure changed. This procedure has been shown not to affect the changes in plasma [K] caused by eipinephrine (2 1). Femoral arterial pressure and lead II ECG were recorded.The adrenergic a-receptor antagonist phenoxybenzamine HCl was administered (5 mg/kg) in 95 c/c ethanol solution (50 mg/ml), and the adrenergic, &receptor antagonist propranolol HCl was administered (3 rng/kg) in aqueous solution (30 mg,/ml) by intravenous infusion during a lo-min period. The infusion of each of the vehicles alone at this rate (0.01 ml/kg per min) produced no detectable effects (35). The interval between the beginning of the infusion and the administration of cpincphrine was: phenoxybenzamine, 60 min, propranolol, 30 min. When both antagonists were given tQ the same animal, phenoxybenzaminc was administered before propranolol. The interval between the infusion of each antagonist and epinephrine was the same as given above.
