A rapid and convenient technique for the simultaneous high-yield purification of multiple bands of DNA fragments from a single agarose or polyacrylamide gel is described. This technique utilizes the best features of several previously published methods, including electrophoretic transfer of DNA to DEAE-cellulose paper, transfer at right angles to the gel surface to yield a replica of DNA bands on the paper, and use of simple handmade microfilters to recover the DNA. The use of a new device, the electrophoretic transfer chamber (e.g., the Trans-Blot apparatus made by Bio-Rad), permits the incorporation of these desirable features into a single method and simplifies the manipulations involved.
