Mouse skin application has been used for the determination of car-cinogenicity of oils, tars and chemicals for more than 60 years [16, 20, 27, 28]. The use of this test system led to the isolation, characterization and synthesis of many aromatic hydrocarbons which constitute an important group of chemical carcinogens [7]. This test system is used in many laboratories in the pursuit of certain aspects of chemical carcinogenesis dis-cussed in this review [11, 25, 31]. The reasons for the continued use of this model center around its value in chemical carcinogenesis studies. In addition, the ease of administration of test materials, accurate and quan-titative observations of rate of tumor appearance, tumor multiplicity, irri-tancy and other toxic effects are important factors in deciding on this test system.Of laboratory animals most commonly used, ie, mice, rats, rabbits and guinea pigs, mice are the most susceptible to skin tumor induction by chemical agents. The tumors induced are benign papillomas which may regress or progress to squamous carcinomas. The latter are commonly encountered in humans; melanomas, which are at times fatal in humans [1], rarely occur in mice.
