Dendritic cells are the most potent stimulators of antigenspecific immune responses including antitumour responses. 1–3 Potent immunostimulatory dendritic cells can be isolated from peripheral blood or cultured from circulating precursor cells. 1–4 The safety and efficacy of these cells is now being tested in clinical trials; data from skin, prostate, and B-lymphocyte have already been published. 1–3We did a pilot study of cultured blood dendritic cells in patients with metastatic renal-cell carcinoma5 according to a protocol approved by the local ethical committee. Patients receive at least three intravenous infusions of activated, CD83+ dendritic cells4 simultaneously charged with autologous tumour cell lysate and with the immunogenic protein, keyhole-limpet haemocyanin (KLH). 5–10106 dendritic cells (microbiologically tested for sterility) were given per infusion. Four patients have been enrolled in the study protocol and treatment has been completed in two.
