The mechanism of a-galactosi-dase action was studied kinetically. A comparison of the rate of their enzymic hydrolysis shows 6-bromo-2-naphthyl>1-naphthyl->2-naph-thyl-[alpha]-D-galactopyranosides. Such data suggest that the influence of the aglycone group in the [alpha]-D-galactosides is that of a steric acceleration. Km and Vmax for the 3 naphthyl [alpha]-D-galactopyranosides were obtained. Mercuric acetate was found to be an inhibitor for [alpha]-galacto sidase whereas galactose, glucose, fructose, lactose, galacturonic acid, cystelne, histidine, imidazole, and aniline were found to be noninhibi-tors. Certain metal cyanides were found to be activators.
