mplified fragment length polymorphisms (AFLPs) produced with EcoRI/MseI and PstI/MseI restriction enzymes and single sequence repeats (SSRs) were used for constructing molecular map based on 129 F 2 individuals obtained from the crossing of two unrelated sweet sorghum inbred lines (LP29/1 and LP113A). Eight pairs of primers were tested and 140 clear AFLP polymorphic fragments were amplified between the parental lines. A mean of 20 and 32.7 polymorphisms were obtained for the FcoRI/MseI and PstI/MseI primer combinations, respectively. One hundred three fragments segregated according to the Mendelian distribution and 94 (57 PstI/MseI and 3- EcoRI/MseI were significantly linked to the framework map, forming 10 major linkage groups spanning 1381.5 cM. Fifteen SSR markers with known location on reference linkage groups were mapped onto the AFLP linkage map.The comprehensive map includes 144 markers and spans 1659.9 cM, while eight markers retrained unlinked. The framework map was used to identify quantitative trait loci on 99 F 3 progenies, each tracing back to a single F 2 plant. The following traits were investigated: sugar percentage, sucrose percentage, dry matter yield, sugar yield, flowering time, plant height and germination ability at low temperatures. Simple (SIM) and composite interval mapping (CIM) were used for QTL analysis. Significant QTLs were found for all the traits investigated, except for sugar percentage, indicating that there was no segregation of genes having major effects on this trait in the population investigated.
