Nitric Oxide participates in various physiological processes and the lack of in vivo measurement tools limits the understanding of its function in plant. In this paper, a poly Ni(II)-tetrasulfonated phthalocyanine/poly(o-phenylenediamine)/Nafion modified graphite electrode for NO detection was constructed and characterized by cyclic voltammetry. This modified electrode showed good sensitivity in both phosphate buffer saline (PBS) solution (1.18 nA/mu M) and 10% tobacco leaves homogenate (1.09 nA/mu M) with a high selectivity against the interferences from ascorbic acid (AA), sodium nitrite (NaNO2) and hydrogen peroxide (H2O2) due to the combination of poly(o-phenylenediamine) (PPD) and Nafion coatings. The continuous in vivo monitoring of NO in plant was accomplished using this NO sensor by amperometry on the tobacco leaves with ultraviolet C radiation as stressor. This NO sensor shows a great potential to facilitate the NO research in plant.
