Efforts to induce the formation of pemphigus antibodies have led several research groups to conduct rabbit immunization experiments. These have induced antibodies to the intercellular areas of stratified epithelia as detected by the indirect immunofiuorescence (IF) te~ t.'.~.'.''However, none of these demonstrated in vivo bound intercellular antibodies, thus suggesting that antibodies produced were pemphigus-like. These antibodies yield indirect IF staining patterns resembling those of pemphigus antibodies. From the standpoint of the antibody producer. the most important difference is that true pemphigus antibodies bind in vivo to their antigens in the normal skin and mucosa and produce epithelial damage whereas pemphigus-like antibodies fail to doThe reports of Michel, Schiltz, and their co-worker~~~~ on the binding of pernphigus antibodies to skin explants with subsequent development of intraepithelial lesions in the absence of complement are the most convincing experimental evidence for the pathogenic potential of pemphigus antibodies. Recently, we have adapted this organ culture system for use with mucosal explants. The purpose of this study was to elicit antibodies to the intercellular antigens of stratified epithelia by immunization of monkeys with a saline extract of human esophageal mucosa containing pemphigus antigen. We also characterized these antibodies with regard to their pathogenic potential in organ culture. Because a number of reports describe cases of human pemphigus provoked by D-penicillamine, some of the animals were pretreated with this drug.'.'*
