A mouse ubiquitin clone was used at low stringency to identifyT. brucei bruceiubiquitin sequences from a cosmid library (constructed in the vector c2×B). From a positive recombinant cosmid clone (pJL4) a ubiquitin-hybridizing 228bpSalI fragment (pUS02) was cloned. Complete sequencing of the pUS02 insert showed it to possess 80% identity and 94.7% amino acid identity with the mouse ubiquitin, confirming its selection as a ubiquitin gene. Hybridization of radiolabelled, clonedT. brucei bruceiubiquitin DNA to southern blots ofT. brucei bruceigenomic DNA revealed 4 regions of sequence identity. The primary structure of the most strongly hybridizing genomic band is presented. It contains up to 30 ubiquitin coding repeats. The primary organization of theT. brucei bruceipolyubiquitin gene resembles that of yeast and other organisms, rather than that of the relatedT. cruzi.InT. brucei bruceithe polyubiquitin carboxy terminus is a single, non-ubiquitin amino acid extension.
