Snakeheadfishvesiculovirus(SHVV),amemberofthefamilyRhabdoviridae,hascausedmass.mortalityinsnakeheadfishcultureinChina.PrevioustranscriptomicsequencingofSHVV-infected.andnon-infectedstripedsnakeheadfishcells(SSN-1)showedthatglutaminase (GLS),thecritical.enzymeofglutaminemetabolism,wasupregulateduponSHVVinfection.Itthereforedrewour.attentiontoinvestigatingtheroleofglutamineinSHVVpropagation.Glutaminedeprivation.significantlyreducedtheexpressionofthemRNAsandproteinsofSHVV,andtheproductionof.virusparticles,indicatingthatglutaminewasrequiredforSHVVpropagation.Glutaminecanbe.convertedtoglutamatebyGLS,andthenbeconvertedtoa-ketoglutarate, tojoininthetricarboxylic.acid(TCA)cycle.AdditionoftheTCAcycleintermediatea-ketoglutarate,oxaloaceticacidor.pyruvatesignificantlyrestoredSHVVpropagation,indicatingthattherequirement ofglutaminefor.SHVVpropagationwasduetoitsreplenishment oftheTCAcycle.InhibitingtheactivityofGLSin.SSN-1cellsbyaninhibitor,bis-2-(5-phenylacetamido-1,3,4-thiadiazol-2-yl)ethyl sulfide,decreased.SHVVpropagation,whileoverexpression ofGLSincreasedSHVVpropagation.Takentogether,our.datahaverevealedtherelationshipbetweenglutaminemetabolismandSHVVpropagation.
