APLASTIC ANAEMIA IN SUBLETHALLY IRRADIATED MICE GIVEN ALLOGENEIC LYMPH NODE CELLS

When 10,000,000 lymph node cells from CoH/H mice were injected intravenously into CBA/H mice on the day after a sublethal exposure to 450 or 600 rads, about 1/2 the mice developed a fatal aplastic anemia. Deaths from anemia occurred between 13 and 142 days later with few of the ordinarily accepted signs of a graft-host reaction. The marrow was grossly aplastic. Hematologi-cal and histological observations after 600 rads and experiments with higher midlethal radiation doses and mixtures of C3H or CBA lymph node or bone marrow cells showed that the allogeneic lymph node cells specifically prevented regeneration of host-type bone marrow. The results may indicate how somatic mutation in immunologically competent cells could result in the sporadic occurrence of aplastic anemia in man.
