The influence of cytochalasin B (CB) on TA3 ascites [mouse mammary] tumor cells was in vivo to assess whether CB induced cell paralysis would affect the transplantation behavior of the cells and in particular tumor distribution after i.v. cell infusion (experimental metastases). Tumor cell pretreatment with CB (1 .mu.g/ml) did not alter the s.c. or i.v. transplantability of TA3 cells. Pretreatment with 10 .mu.g/ml CB, consistently increased the incidence and number of extrapulmonary tumor takes from i.v. transfused cells. The amount of pulmonary tumors was not significantly altered. S.c. transplantability was not affected by 10 .mu.g/ml CB. The importance of cell mobility and cell surface topography for tumor cell nidation in vessels is discussed.
