After more than two decades of virtual stagnation, progress has recently been made in understanding the pharmacology and function of 5-hydroxytryptamine (5-HT) receptor subtypes. This has largely resulted from the use of new selective potent agonist and antagonist drugs that have now become generally available. Three basic subtypes of 5-HT receptors have been shown to exist — 5-HT1, 5-HT2and 5-HT3— and evidence is emerging to suggest heterogeneity within the 5-HT1and 5-HT3receptor categories. Here we review recent developments in this field with particular reference to the 5-HT3receptor, which is widely distributed throughout the peripheral nervous system where it mediates many of the excitatory actions of 5-HT.
