MethodsPreparation of Transcarboxylase and the Subunits. Transcarboxylase was isolated from P. shermanii (52W strain) and assayed by the method of Wood et al.(1969). The outer subunit complexed with two biotinyl subunits (designated 6S subunit) and the central subunit in hexameric form were isolated by stepwise dissociation of transcarboxylase bound to a cellulose phosphate column (Bahler et al., 1981). The isolated subunits were found to be homogeneous as evidenced by SDS-PAGE.
