C eramidase is a hydrolase capable of cleaving the N-acyl linkage between a sphingosine base and a fatty acid of ceramide. Recent extensive studies have revealed that ceramidases can be classified into three different families; acid, neutral and alkaline enzymes, based on not only their catalytic pH optima but also their primary structure. The genetic information of neutral ceramidases is highly conserved from bacteria to humans. This Chapter summarizes recent progress in the study of neutral ceramidases.
