The humoral antibody responses to reciprocal skin homografts between two inbred strains of rat differed considerably in timing and in degree. The Lewis recipients developed cytotoxic antibody by the sixth day after grafting, but the titers achieved were never high and declined to low levels by the 26th day. BN recipients, on the other hand, did not show circulating antibodies on the sixth day, but then developed substantially higher antibody levels which continued to rise throughout the 26-day observation period. Over a wide range, the size of the skin graft bore little relation to the titer of antibody achieved, Massive skin homografts exhibited paradoxically prolonged survival, in spite of serological evidence of an active immune response. The data are consistent with the hypothesis that some specific factor other than the cytotoxic antibody of serum is the limiting factor responsible for the long survival of massive homografts.
