The association of osteoporosis with the post-menopausal state and the belief that estrogen replacement may be efficacious in treating the osteoporotic process has created considerable interest in the action of estrogens on bone. The purpose of the present study was to ascertain whether specific receptor sites for estradiol could be demonstrated in rat bone, thus providing a basis for the direct action of estrogen at the bone cell level. Cytoplasmic extracts from a variety of bones were incubated with [3H]estradiol and bound receptors were measured by Sephadex chromatography. Cultured bone cells from fetal calvaria and cytosol from adult ovariectomized females failed to exhibit estrogen receptors, although glucocorticoid receptors could be demonstrated as previously described. Mixing experiments indicated that the absence of receptors was not due to interference by the other factors in bone cytosol or by attachment of receptors to bone hydroxylapatite.
