Doxorubicin (DXR), or adriamycin, is an antitumoral antibiotic, obtained by the aerobic fermentation of Streptomyces peucetius var caesius or by chemical synthesis from daunomycin (Arcamone et al., 1972). The drug molecule displays two different chemical domains: adriamycinone (showing an anthracycline ring structure) and dannosamine (an amino sugar presenting a single free amino group) which are botmd together by a glycosidic bond.DXR exhibits a significant antitumor activity (Di Marco et al., 1969; Tan et al., 1973; Bloomfield et al., 1972; O'Brian et aI., 1973; Goldin et al., 1975) and in particular plays a central role in the chemotherapeutic management of acute myelogenous leukemia, non-Hodgkin's lymphomas, and carcinomas of the breast, lung and thyroid (Young et al., 1981). The clinical use of high doses of DXR (exceeding 500 mg/ma body surface area) is strongly compromised by the development of an irreversible congestive cardiomyopathy and heart failure in human patients (Lefrak et al., 1973). This heart injury is dose-dependent and related to the total cumulative dose of the administered drug (Henderson and Frei, 1979).
