Using a new reagent,103ruthenium red, glycosaminoglycans can be detected on cellulose acetate electrophoretograms at much greater sensitivity than with other methods currently available. Nonradioactive ruthenium red has been used extensively in the past as an electron microscopic stain for polyanions. Commercially available103RuCl3can be easily converted to103ruthenium red and this beta-particle-emitting isotope can be detected with autoradiography. The limit of detection for heparin, heparan sulfate, or chondroitin sulfate is 2–4 ng.
