Poly(A)+ RNA was isolated from maize by affinity chromatography on columns of oligo(dT)-cellulose. Information concerning the nature of the nucleotides surrounding N6-methyladenosine was obtained by radioactive labeling in vitro of the 5''-termini of oligonucleotides liberated by digestion of the RNA with base-specific RNases. The major sequence containing N6-methyladenosine was Pu-m6 A-C.
