The hypothesis that the formation of the receptor estrogen complex is a requirement for the induction of uterotrophic responses was tested. A single injection of 2.5 μg of estradiol given to immature rats results in the rapid accumulation of the receptor estrogen complex by the nuclear fraction of rat uteri. However, a second injection at 3 hr following the initial injection, when the amount of cytoplasmic receptor is very low, does not elevate nuclear receptor content. If the estrogen receptor is causally related to uterine sensitivity to estradiol one would expect that uterine responses would also be insensitive to estrogen at 3 hr after estradiol treatment. To examine this possibility, rats which were pretreated with estradiol for 3 hr received an additional estradiol injection and the extent of short (wet weight and glucose oxidation) and long term (wet and dry weight) responses was determined.
