The properties of a RNase inhibitor found in the supernatant fraction prepared from rat liver homogenates were investigated. The inhibitor, which is heat labile and non-dialyzable, is active against crystalline pancreatic RNase and alkaline RNases from rat tissues, but apparently has little effect on acid RNase. The distribution of inhibitor in the supernatant fractions prepared from several rat tissues was determined. Brain, liver and lung contain the highest concentrations in de-creasing order, while kidney and muscle contain smaller amounts. Large quantities of RNase are normally bound to the inhibitor. The bound RNase is equal to or greater than the total mitochondrial alkaline RNase activity of liver or kidney. The bound enzyme may be released and the inhibitor inactivated by Pb++ and p-chloro-mercuribenzoic acid but not by iodoacetate or iodoacetamide.
