METHODSThe subjects of this study were II normal volunteers, ages 2 r-29, resident at the Clinical Center, National Institutes of Health. Each gave a history of normal taste and smell. None had experienced significant injury or infection in the oral or pharyngeal areas. Each had normal dentition, occlusion, palatal shape and physiognomy determined by inspection and by anterior and lateral cephalometric radiographs. Fungiform and circumvallate papillae were present on the tongue of each subject. Uvular and gag reflexes were present in each subject.Studies were carried out with the subject seated comfortably in a standard dental chair in an air-conditioned room in the Dental Clinic of the Clinical Center. All studies were performed between 2: oo and 5: oo PM in order to obtain maximal taste sensitivity in each subject (7). Subjects did not smoke or eat for at least I hr prior to the test. Initially the subject was familiarized with the experimental procedure, after which detection and recognition thresholds for NaCl for salt, sucrose for sweet, HCl for sour, and urea for bitter were obtained with a forced, three-choice technique, carried out by methods previously described in detail (7). After these thresholds were determined, either the tongue or palate was anesthetized. The tongue was anesethtized by bilateral injection of 1-3 ml of 2% lidocaine HCl with I/I oo, ooo epinephrine into the pterygomandibular space, so that the entire tongue surface, including areas innervated by the lingual and glossopharyngeal nerves, was anesthetized.
