TRANSCRIPTIONAL ENHANCERS were discovered in eukaryotic organisms. They were originally defined as cis-act-ing DNA sequences involved in activation of transcription by RNA polymerase II 03) and were characterized by the fact that they functioned efficiently at large distances from the promoters that they regulated, both downstream and upstream of these promoters, and in either orientation~. Transcriptional enhancers are now known to be binding sites for proteins and to be involved in regulating transcription in both a negative and positive manner. Tra,-scrlptional enhancers and enhancerbinding proteins play critical roles in controlling the differentiation and development of eukaryotic organisms and in regulating their metabolism-3. There has therefore been considerable interest in how enhancers communicate with their cognate promoters and influence promoter function. The precise target of a eukaryotic enhancerbinding protein has not been determined in any case.
