By means of immunohistochemical .alpha.-amylase determination techniques and Calcofluor white M2R New staining of residual endosperm cell walls it was possible to observe transport of .alpha.-amylase and breakdown of cell wall material in germinating seeds of H. vulgare cv. Nordal. When barley seeds, of which the distal tip had been removed, were treated with GA3 the importance of the distal aleurone cells in the production of both .alpha.-amylase and cell wall breakdown hydrolases was clearly seen.
