Spermatophore formation in argasid ticks was studied by interrupting mating (i.e. removing the female from the male at the very beginning of copulation), and by interrupting spermatophore formation (i.e. by removing various components of the spermatophore as soon as they appeared on the genital aperture of the male). The spermatophore is built step by step starting with the outermost layer. All components are ejected one after another in a well defined manner. Most components are each secreted into the existing layers and then assumes its correct position. The relationship between the various secretions and the components of the spermatophore was studied by histochemical methods.
