Dynamic changes in the monolayer permeability and rearrangement microfilaments in ECV304 cells after Dengue virus infection

To investigate the real-time rearrangement of microfilaments andalteration in the monolayer permeability in endothelium-like cells five days after Dengue virus infection,immunofluorescence assay was applied to observe the rearrangement of microfilaments in ECV304 cells and human umbilical vein endothelial cells(HUVEC),and the transwell model was used to investigate the alteration of the monolayer permeability in ECV304 cells.The virus titer was measured by plaque assay.It was found that rearrangement of the microfilaments could be demonstrated both in HUVEC and ECV304 cells one hour after Dengue virus infection,and peak of increased monolayer permeability occurred at the third day after infection in consistent with the top of virus titer and the dramatic decrease of the stress fibers.It suggests that the increase in monolayer permeability in ECV304 cells might be associated with the rearrangement of microfilaments induced by the high titered virus in these cells.
