Although echinoderm eggs have been used by many investigators in studies of cell division, especially cytokinesis and physico-chemical properties of the mitotic apparatus, chromosome movement has scarcely been investigated in these materials because chromosomes were hardly observable in the living state by conventional light microscopy. Recently, Hiramoto et al.(1) found that chromsomes can be observed in living eggs of the sand-dollar, Clypeaster japonicus with a differential interference microscope. This finding has enabled us to use this material for experimental studies of the mechanism of chromosome movement in the living state. In the present study, we have analyzed the location where the motive force for anaphase chromosome movement is generated by means of micro-operations on the mitotic apparatus and by destroying a pre-selected region of the mitotic apparatus by local application of Demecolcine.
