While there is evidence that some variegating loci are inactivated as a consequence of DNA loss, due to differential replication or DNA elimination 13, 19, it is clear that in many cases of PEV this does not occur 2 m. Inactivation is generally the result of changes in chromatin structure'v'6. One would therefore expect mutations that modify the extent of inactivation at variegating loci to lie in genes encoding chromosomal proteins, their modifiers, or assembly factors (reviewed in Ref. 8). Many of these mutations have been identifieda2, 23, including both suppressors of PEV [Su (var)], which suppress inactivation of the variegating gene, and znhancers of PEV [En (var)], whicil enhance inactivation. Mutations at as many as 120 loci can have a dominant effect on PEV 2"~. Several such modifier loci have been cloned and characterized (reviewed in Refs 8, 25). Interestingly, many
