The pulmonary damage of paraquat dichloride (PQCl2) is believed to be mediated in part by superoxide anion. The effect of parenterally administered superoxide dismutase (SOD) on PQCl2toxicity was studied in two species. Mortality did not decrease in mice given SOD at 10 or 35 mg/kg of intermittent parenteral injections after 25 mg/kg iv PQCl2(45 to 65%) compared to controls (55%). Similarly, rats given SOD intermittently after po PQCl2showed no significant reduction in mortality (50% versus 65%) or in time to death. Adolescent rats administered parenteral PQCl2followed by intermittent parenteral SOD had the same death rates as rats receiving only the PQCl2. Since SOD has a short half-life, the effects of 5-day continuous iv infusion of high and low doses of SOD on rats given increasing doses of parenteral PQCl2were studied. No protective effect of SOD was noted under any of these conditions. No measurable level of exogenous SOD was found in rat lung after 2 days of continuous infusion. The previously reported protective effects of SOD on PQCl2toxicity in rats could not be confirmed.
