Materials and MethodsFn was purified from fresh-frozenhuman plasma, obtained from the Blood Center of Southeastern Wisconsin, on a ge-latin-Sepharose 4B affinity column (Engvall & Ruoslahti, 1977). The integrity and purity of the protein, before and after labeling with the sulfhydryl reagents, were routinely examined with sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The fluorescence probes 7V-(l-pyrenyl) male-imide (P), 7-(diethylamino)-3-(4,-maleimidylphenyl)-4-methylcoumarin (C), and fluorescein 5-maleimide (F) were purchased from Molecular Probes (Eugene, OR). Polystyrene latex beads with surface carboxyl groups were purchased from Polysciences (Warrington, PA). The beads used in this study had a diameter of 0.5¿ tm.
