HeLa cells, L cells, human embryonic lung cells, and chick embryos were allowed to grow in the presence of methionine-methyl-14C. The DNA from these cells was then isolated and analyzed for methylated bases by measurement of radioactivity. The only methylated base, other than thymine, found in DNA from L cells and chick embryos was 5-methylcytosine. DNA from human embryonic lung cells contains 5-methylcytosine and a relatively small amount of 3-methylcytosine. HeLa DNA contains these two methylated cytosines plus small quantities of 1-methylguanine, 7-methylguanine, N2-methylguanine, and N2-dimethylguanine. In addition, DNA from HeLa and L cells contains an unidentified methylated component which can be removed from the DNA by treatment with pronase. This unknown substance is not present in DNA from chick embryos and human embryonic lung cells.
