Materials and MethodsThe materials used were as described in the preceding paper (Lillehaug and Kleppe, 1975). The standard assay mixture contained 60 mM Tris (pH 8.0), 9 mM MgCla, 15 mM/3-mercaptoethanol, 0.066 m [-32] ATP, 0.23 mM micrococcal nuclease treated calf-thymus DNA, and ap-proximately 1-1.7 units/ml of T4 polynucleotide kinase. The normal incubation time was 30 min at 37 if not other-wise stated. The other methods used are described in the preceding paper (Lillehaug and Kleppe, 1975).
