The activity of enzymes that labilize known CH bonds can be estimated in intact tissue by following the loss to water of tritium from specifically labeled substrates. The accumulated tritium oxide (3HHO) in the medium is recovered by direct sublimation of aliquots of the medium into scintillation vials and the radio-activity measured by liquid scintillation spectrometry.
