The cells which comprise the mononuclear phagocyte system (MPS) are widely distributed throughout the body under steady-state conditions and are recruited in increased numbers following injury, infection and immune stimulation. Their properties vary a great deal depending on their fUnctional state and on local interactions with cells and extracellular molecules in their microenvironment. Recent studies have explored the cell biology and biochemical properties of macrophages and have begun to define the relevance of their activities to a range of disease processes. This review will outline some of the background work that has led to an appreciation of the diverse properties which macrophages display within the body. As a result of these studies we have broadened our view of the MPS to include physiologic homeostatic functions in addition to their traditional role in host defence. We shall refer to recent publications which illustrate the nature and functions of macrophages in tissues and cite appropriate reviews. Other chapters in this volume deal with aspects of macrophage heterogeneity within the intact host.
