A water-insoluble, self-standing cast film of a DNA-lipid complex was prepared by mixing both aqueous solutions of anionic DNAsand cationic amphiphiles at a charge ratio of 1: 1. Intercalation behaviors of dye molecules into a cast film of a DNA-lipid complex deposited on a quartz-crystal microbalance (QCM) was followed quantitatively by observing frequency changes of a QCM in an aqueous solution.DNA molecules are of interest as a molecular recognition site such as specific intercalation behaviors of dye mol-ecules between base-pairs. Intercalation of dyes has been observed by spectra changes of UV and/or visible absorptions of dyes due to the penetration into the hydro-phobic base pairs2, 3 by X-ray4 and NMR5 6789investigations and by viscosity changes of elongated DNA molecules in aqueous solutions. Theseconventional methods have some difficulties in detecting directly and quantitatively what amount of dye molecules are intercalated into DNAs. In this paper, we prepare water-insoluble, self-standing cast films of a 1: 1 DNA-lipid complex by mixing DNA polyanions and cationic dialkyl amphiphiles. The DNA-
