The use of trimethylsilyl (TMS) derivatives in mass spectrometry was first introduced a decade ago by Sharkey, Friedel, and Langer (7), primarily for the analysis of mixtures of alcohols (2). Since that time TMS derivatives have found wide application in gas-liquid chromatography—eg, 3-6, be-cause of their ease of preparation, thermal stability, and volatility. In recent years, renewed interest in the mass spectra of these compounds—eg, 1, 2, 7-16 has come about primarily for two reasons. First, the advent of combination gas chromatography-mass spectrometry has provided a logical extension of gas chromatographic studies to include the mass spectrometry of many TMS derivatives of natural products. This has been accomplished most successfully in the case of sterol and bile acid derivatives (75). Second, the enhanced volatility of TMS derivatives has provided a powerful means of extending the scope of mass spectrometry to include polyfunctional compounds of very high molecular weight—eg, 7, 14.
