Steady-state concentrations of individual cytoplasmic poly (A) RNAs were determined by hybridization to cloned double-stranded cDNA fragments on nitrocellulose filters. The DNA excess hybridization reaction and X-ray film exposure conditions were properly controlled to allow quantitative analysis of the autoradiograms. mRNA populations from Kc tissue culture cells, embryos, third-instar larvae, pupae, and adults ofDrosophila melanogasterwere analyzed. Our results show that different stages in development exhibit distinct mRNA populations with respect to 20 genes employed in this study. This is due to individual mRNAs which showed characteristic changes during development reflecting the fluctuations in mRNA complexity or abundance.
