Replication of the circular chromosome of Escherichia coli (Cairns 1964) is bidirectional (Masters and Broda 1971; Bird et al. 1972; Prescott and Kuempel 1972). Replication appears to be initiated at a fixed site (Marsh and Worcel 1977) which has been located about 20 megadaltons (MD) counterclockwise of the ilv operon (von Meyenburg et al. 1977) at 82.5 minutes on the genetic map (Bachmann et al. 1976). For this site, the origin of replication of the chromosome, the symbol oriC has been introduced (Hiraga 1976). Here we review our approach to the genetic mapping of oriC through isolation of specialized transducing phages Aasn carrying oriC and their establishment as minichromosomes (von Meyenburg et al. 1978). We describe the construction of minichromosomes consisting exclusively of E. coli chromosomal DNA. In addition, we discuss initial attempts to characterize the in vivo replication and partitioning of minichromosomes, as well as their effects on the growth physiology of the host.
