Physiol. 226 (s): 1227-1231. 1974.~ Pyruvate kinase and lactate dehydrogenase were measured in some or all of the following structures of rat kidney from late fetal age to the adult: glomeruli, proximal, and distal convoluted and straight tubules, thin-limb areas of the medulla, the neogenic zone, mesenchyme, and medullary ray. Samples weighing 0.01-l pg were dissected for analysis. Pyruvate kinase ranged MO-fold in the adult kidney from the proximal convoluted tubules (lowest), the glomeruli and proximal straight tubules (intermediate), the distal tubules (high), to the thin-limb area (highest). The ratio of lactate dehydrogenase to pyruvate kinase varied from 15 in proximal convoluted tubules to 0.3 in thin-limb areas. At all ages the activity of pyruvate kinase at low P-pyruvate concentration was stimulated by fructose diphosphate in proximal tubules but not in other structures. During gluconeogenesis glycolysis would be wasteful. Gluconeogenesis is probably restricted to the proximal tubules. It therefore seems significant that pyruvate kinase is not only low in proximal tubules, but also susceptible to allosteric control. renal tubules; development of nephrons; allosteric pyruvate kinase
