An X-ray crystal analysis of qo-lactone N-{1-{N-{4-{[3-hydroxy-5-methyl-1-oxo-4-(NL-threonylamino) heptyl]-oxy}-2, 5-dimethyl-l, 3-dioxohexyl}-L-leucyl}-L-prolyl}-N, O-dimethyl-L-tyrosine hydrobromide hydrate (la), C42H66N501~-. Br-. H20, was obtained in order to de-termine the backbone folding of the macrocycle and to compare the results obtained with those reported pre-viously for the natural product didemnin B (lb). Some differences were noted in the torsion angles of the two conformers of the hydrobromide salt, denoted (la) and (la'). The conformation of (la') resembled the confor-mation of (lb) more closely than did that of (la). Cer-tain regions of both crystal backbones were more flexi-ble than those in didemnin B; however, the transannular hydrogen bonds in both (la) and (la') were somewhat stronger than in (lb).
