From A. castellanii an actin gene was isolated and completely sequenced which is interrupted by a 129 base-pair intervening sequence following the codon for amino acid 105. S1 nuclease mapping experiments indicated that this gene (actin gene I) is expressed in vivo. There is only one size class (1300 nucleotides) of actin mRNA in Acanthamoeba. Specific fragments of the isolated gene were used to demonstrate that there are at least 3 and possibly more actin genes in this organism. As deduced from the DNA structure, the Acanthamoeba actin I is a protein of 374 amino acids and has an unusual Gly residue at its N-terminus.
