Electron Microscopic and Biochemical Study of the Neutrophilic Granules from Leucocytes

The neutrophilic leucocytes were collected from inflammatory exudates induced in rabbits by intraperitoneal injection of sterile saline. Neutrophilic granules as well as other subcellular fractions were separeted by differential centrifugation. Each fraction was observed in an electron microscope, by means of both thin sectioning and negative staining methods. The neutrophilic granule fraction which consisted of less dense granules, ie. A-granules, was almost homogeneous in size (0.3 μ. diameter) and density. Contamination of mitochondria and other cellular components was scarecely observed. The limitting membrane of the granule has double-membrane structure. Inside of the membrane was generally filled with microgranules of 300 Å in diameter. Occasionally, lamellar structuer was observed in the neutrophilic granules. This fraction contains a new cytochrome which was designated as cytochrome B (558, neutrophils) by the authors. On the other hand, the “mitochondrial” fraction contained very small amount of mitochondria as revealed by both electron microscopy and spectrophotometry. This fraction was mainly composed of the dense neutrophilic granules, B-granules, in which the presence -of a new haemoprotein was suggested spectrophoto-metrically, and a tentative nomenclature, haemoprotein 560 (neutrophils) was proposed for this haemoprotein.
