To establish a dose of digoxin for beneficial plasma concentrations in budgerigars, 15 of 25 mature sparrows of mean weight 31 g were given 0.625 mg/kg digoxin and 10 were given 0.3125 mg/kg orally as a tincture containing 0.05 mg/ml. Blood samples from one fifth of the birds at each of 5 intervals over 24 hours showed that half-lives of these doses of digoxin were 26 and 21 hours, respectively, indicating that digoxin given once daily would maintain plasma concentrations. Three of 9 mature budgerigars of mean weight 28 g were given orally 1 of 3 daily doses for 5 days: 0.36, 0.18, 0.09 mg/kg. Digoxin concentrations at 12 hours after the fifth dose were 12.8, 7.6 and 4.8 ng/ml plasma. Based on this, 4 budgerigars were given 0.02 mg/kg/day for 5 days. Mean plasma concentration was 1.6 ng/ml, considered to be within the therapeutic range in mammals, although the recommended dosage varies inversely to mass, so 0.02 mg/kg is lower than that estimated. Thus to provide a daily dose of 0.0005 mg, 0.05 ml of a 1:4 dilution of the tincture is required.
