[beta]-Cyanoalanine, the neurotoxin naturally present in various species of Vicia (vetch), is shown to be an effective inhibitor in vitro of rat liver cystathionase. Inhibition was reversible with increasing amounts of substrate, and no evidence of an inhibitor-pyridoxal phosphate interaction was found. Structure-activity studies suggest that the cyano group and the N-unsubstituted amino acid of L configuration are probably essential structural features of the inhibition.
