The initial objective of these experiments with the isolated, perfused animal liver was to achieve a reproducible experimental preparation that would function as a normal liver. The results reported as to flow, oxygen utilization and parameters of liver function indicate that this has been accomplished. Because of the initial difficulties encountered, particularly in the dog, by us and by other workers, it seemed worth while to report the technique in detail. Causes of "outflow block" and unsatisfactory perfusions are too numerous to describe in detail. Meticulous technique and a smooth rapid transition from the in vivo liver to the isolated perfused preparation are essential. Even brief periods of anoxia increase the portal bed resistance and perfusion through the portal vein alone of blood of high oxygen saturation will not establish a satisfactory perfusion. A normal pCO2 is necessary to prevent increasing resistance in the arterial bed and progressive hypoxia, and pH must be maintained between 7.2 and 7.5. Detailed studies on the effects of hypoxia, alterations in acid-base balance and other metabolic changes on flow rates and liver function are in progress. || ABSTRACT AUTHORS: Authors
