A significant body of circumstantial evidence suggests that Ca 2+ and calmodulin are important regulators of cell growth and malignant transformation. The case for such a role is compelling because it is hard to imagine that cell growth and the morphological changes involved in the malignant transformation of cells could progress through the required complexity of accompanying biochemical events without central reliance on Ca 2÷-calmodulin regulation. Specific mechanisms for calmodulin in these processes, however, are far from established.This review will attempt to provide a perspective on the current state of the evidence linking calmodulin to regulation of cell growth and transformation. Many of the arguments pertinent to the multifarious roles of calmodulin in growth and cell function, however, rest on our understanding of calcium and other ions as cofactors and regulators of cellular metabolism. Therefore, to set the scene for a cogent discussion of calmodulin as a central pluripotent regulator of cell function, it is necessary to establish some background for calcium's role relative to other ions as a specific chemical second messenger and to briefly discuss calmodulin's physical and functional properties.
