MATERNAL DIABETES DURING PREGNANCY - CONSEQUENCES FOR THE OFFSPRING

The immediate and long-term consequences for the offspring of maternal diabetes during pregnancy were studied in pregnant rats with induced mild and severe diabetes (first generation). Information was gathered on foetal, neonatal and adult offspring (second generation) and on their adaptation to pregnancy as well as the status of their foetal and adult offspring (third generation). Maternal diabetes during pregnancy induced alteration in foetal metabolism with consequences persisting in later life. Diabetes or a diabetogenic tendency was found to be transmitted from one generation to another. A clear divergence in the influence on foetal and adult offspring of mild versus severe maternal diabetes was observed. Independent of the degree of maternal diabetes or nature of glucose tolerance in the adult offspring, in pregnancy (second generation) they developed mild gestational diabetes. Their foetuses (third generation) therefore developed in an abnormal intrauterine milieu and displayed the typical features of foetuses from mildly diabetic dams. When fully grown they displayed a disturbed glucose tolerance. Although these data do not enable extrapolation to the situation in man they point to the impact of diabetic intrauterine milieu on the developing foetus and confirm the persisting influence on the offspring throughout life.
