为了评估地表水中导致人类急性胃肠炎的三种胃肠炎病毒的存在,在中国北京市开展了一项为期一年的研究。2006年9月至2007年8月,在河流或湖泊中使用全球定位系统确定的9个收集点共采集了108个城市地表水样本。采用HA电负性过滤器对水样进行病毒浓缩,然后对轮状病毒(RV)、星状病毒(AV)和诺如病毒(NV)进行聚合酶链反应(PCR)。结果表明,不同地点水样中检出的病毒数量不同,共检出63种病毒株,其中轮状病毒(48.1%)最多,其次是星状病毒(AV, 5.6%)和诺如病毒(NV, 4.6%)。实时荧光定量PCR检测RV, RV浓度范围为0 ~ 18.27个基因组拷贝·L−1。在寒冷季节(9 ~ 2月),RV在地表水中的分布较为丰富,而在温暖季节(3 ~ 8月),RV的分布较少。本研究中发现的RV的高检出率为RV在北京人群中有一定的传播频率提供了有力的证据。RV水平与粪便大肠菌群(R2= 0.02)和粪肠球菌(R2= 0.02)密度均无统计学意义。我们的研究表明病毒在水生环境中存在时间较长,并强调肠道病毒组是最可靠的环境监测。
To assess the presence of three gastroenteritis viruses responsible for human acute gastroenteritis in surface water, a 1-year study was carried out in the city of Beijing, China. A total of 108 urban surface water samples were collected from nine collection sites which were defined with a global positioning system in rivers or lakes from September 2006 to August 2007. The water samples were subjected to virus concentration using an HA electronegative filter, followed by polymerase chain reaction (PCR) for rotavirus (RV) astrovirus (AV), and norovirus (NV). It showed that the number of viruses detected in water samples from different sites was variable, totaling 63 virus strains, with rotavirus (48.1%) verified as the most prevalent detected, followed by astrovirus (AV, 5.6%), and norovirus (NV, 4.6%). RV was also quantified by real-time PCR and the concentration of RV ranged from 0 to 18.27 genome copies·L−1. And the distributions of RV in surface water were abundant in cold weather (from September to February) while less prevailing in warm weather (from March to August). The high detection rate of RV we encountered in this study provided convincing evidence that RV circulated at a certain frequency in the Beijing population. There was no statistically significant correlation between RV levels and both fecal coliform (R2= 0.02) andEnterococcus faecalis(R2= 0.02) densities. Our study suggests prolonged virus persistence in aquatic environments and emphasizes the enteric virus group as the most reliable for environmental monitoring.
